Apple opens iTunes Store in Hong Kong, Taiwan and 10 other AsiaPacific countries

Many Apple fans on the opposite side of the Pacific from Cupertino haven't had much of a choice to shop from iTunes, even though they've had the App Store for some time. There's now a much better sense of balance: Apple just flicked the switch on the iTunes Store for music and movies in a dozen countries and territories across the Asia-Pacific region. The company singles out our own Richard Lai's Hong Kong as well as Singapore and Taiwan, but we'd be remiss if we didn't mention that Brunei, Cambodia, Laos, Macau, Malaysia, the Philippines, Sri Lanka, Thailand and Vietnam are also getting the media catalogs, which include local content along with the international hits. It's a full catch-up as well, with iTunes in the Cloud re-downloads and iTunes Match subscriptions available in every new country. If you're a huge Andy Lau fan but wanted his albums from the most iPhone-friendly store possible, the wait is over.

Katherine Marshall: Rio +20: Point of No Return?

The United Nations Conference on Sustainable Development (aka Rio+20) is concluded in Rio de Janeiro amidst commentary that ranged from utter despair to very tempered hope. Those who see an existential threat to the survival of the planet and mankind describe the hard won consensus agreements that emerged as pathetically limp. Earth and mankind are at a point of no return when the accelerating pace of climate change becomes irreversible. Point of no return conveys the idea well: it's when an exploratory airplane no longer has enough fuel to return home. The chronic optimists among us see hope in the fact that something did emerge, and indeed that, 20 years after the Rio Earth Summit, the meeting took place. And we hope and wish for coalitions of citizens from around the world with a determination to act.

The Rio meeting was huge. Over 45,000 participated, with delegations from 188 countries plus three observers. Over a hundred heads of state and government participated (with Barack Obama among the absentees). Over 4000 journalists covered the events. The official reports claim $513 billion in commitments and real progress towards a greener, sustainable economy. There were hundreds of commitments to action by companies, NGOs, universities and individuals.

An important story is the large and growing role of religious organizations and leaders among the 45,000. They came from many parts of the world and different faith traditions. Their common message is that the challenges of climate change and care of God's creation represent an urgent moral imperative. Action is critical. The allies include leading global religious and spiritual figures.

The religious voices are not unanimous. Family planning, for example, was an elephant in the room: ignored or deliberately kicked out of the discussion because it still evokes strong reactions mostly from the odd coalition of religious bodies that since 1984 are termed the "unholy alliance": the official Catholic Church, some Muslim voices, and some from the evangelical community. Every suggestion that growing population was a problem that demanded action was contested. Rio provided evidence that family planning is a conversation that needs to be reengaged in religious circles because there is plenty of support as well as opposition.

I participated in a March meeting in the Hague, called "Wings for Rio," that illustrates well the passions and concerns around climate change that brings very different religions leaders together. "Wings for Rio" sought compelling messages that would, like a bird, take wing for the conference. The event was grounded in the 20 year Earth Charter, and driven by Rabbi Arwaham Soetendorp. He was joined by leaders following spiritual traditions, Christian leaders, businessmen determined to restore spirit to the world of commerce, and a diverse group of political allies. Its conclusions took the form of a declaration whose signatories include the Dalai Lama. Its flavor is captured in its concluding phrases:

This is a solemn moment of global decision making. The Rio + 20 Conference provides a historic opportunity to lead the world into a more sustainable future. We do not have another 20 years to lose. To move forward in our best interests and even more in the interests of those yet to be born, we must fundamentally change the concepts that underlie our negotiation practices and realize that only together can we forge inclusive solutions. Humble in the consciousness that the consequences of our decisions and actions will be felt by many generations to come, we turn to the Source of All Blessings for strength and courage. May our children, and our children's children take pride in our actions.

The Hague meeting was a provocative blend of pragmatic talk about green energy and reducing waste and calls to spiritual reflection. We struggled with central challenge: how, morally and still more practically, can we balance the imperative of working to end poverty with recognition that unregulated economic growth can destroy the planet. Moderation is the watchword.

Three interesting voices gave the Hague meeting a distinctive flavor. First, several leaders from indigenous traditions spoke forcefully to the deep respect for nature and awareness of mankind's dependence on the earth that are a thread that binds many traditional religious beliefs and practices. There's plenty to learn from them. The meeting included Bhutan's Minister of Labor who touted the principles and quantitative measures that Bhutan has elaborated as the way ahead for their commitment to a framework of Gross National Happiness (in contrast to Gross National Product). Their hope is that Bhutan's humane and spiritual paradigm will compel leaders in Rio to rethink notions of progress centered on consumption and greed. And 96-year-old Brahma Kumari leader Dadi Janki emphasized repeatedly the wise observation that the place where change must start is within each one of us.

Did these messages resonate in Rio? Probably not in any truly significant way. But they are prophetic in the sense of combining vision with poetry and underlining the ethical arguments that surely everyone must hear. My bet is that the resonance of these spiritual and moral insights will grow as we move forward and that spiritual arguments will compel action at least as powerfully as the convictions of science.

The Definitive Layman's Guide to Diagnosing and Fixing a Slow ...

There are multitudes of web pages written on identifying, locating, and removing viruses, spyware, and adware.? This will be very much a 10,000 foot view of this issue since viruses are ever-changing and answers to specific? virus-related problems can be found, in detail, elsewhere.

Prevention
Prevention is the best way to not have to deal with this problem.? If you are reading this, I assume you already have a virus so I will keep this short.? Know what types of sites you are visiting!? If you frequent known, low risk sites and searches you will have fewer problems.? If you are constantly surfing the seedy underbelly of the web, you will need to learn to recognize risky sites or have some sort of in-browser type virus protection.

Get a good virus protection program. There are many free ones.? My favorites are AVG, Avast, and Microsoft Security Essentials. You only need one.? Norton, MacAfee, and Kapersky are some of the more popular paid virus protection programs. I have two issues with these:

  1. They tend to take control of the root of your system, blocking you from performing tasks on your computer as a means of preventing you from harming the system yourself.? This is not necessarily a bad thing.? If you are new to computing, this can help keep you safe.
  2. They tend to take up lots of resources (memory, disk space, etc).? Sometimes the virus protection program can slow your computer more than a virus would.

If you believe you already have a virus.

1) Start your computer in Safe Mode

  • Reboot your computer and as it is booting up slowly tap the F8 key at the top of your computer (some models use the delete key), about once every 3 seconds or so.?? You should see a black screen that with several boot options on it.?
  • Look for ?Safe Mode with Networking?.?
  • Highlight it by using the up and down arrows, then hit enter. This will load only the essential programs needed to make your computer run (and connect to the internet).?? The screen may look strange since some drivers are not running.? Safe mode should also be showing in the corners of your screen.
  • Once at the desktop, look in your task manager to see what programs are running.? Does anything look strange? Stop any unnecessary programs.

2. Download, Install, and Run HijackThis
Hijackthis from TrendMicro will help remove files that have ?hijacked? your computer.? These files can prevent your virus software from doing its job effectively.? If you are able to get to the internet, download, install, and run HijackThis.? If you are not able to get to the internet, it may be time to call a professional.

  • Download and install HijackThis
  • Run the program
  • Look at each line. Look for the names of companies that you recognize.? Companies like Microsoft, Yahoo, HP, or Norton.? These files are usually safe
  • Look at any lines that do not contain these company names.? Do you recognize the program that is referenced in the line?
  • If you find a line that doesn?t have a familiar program name in it, copy everything after the last ?/?.? This will be the actual file name, usually ending in ?.exe? (executable).
  • Paste that into a web search at Google or Bing or your favorite search engine.
  • Read about the file. Is it safe?? Often it is easy to realize that a file is bad because the top search results will contain words like ?virus? or ?how to remove?.
  • Once you are certain that the file is bad, put a checkmark in the box next to it in HijackThis
  • Once you have all of the bad lines checked, click ?Fix Checked?
  • If you are not sure, do another scan and this time save a log file.? This file can be uploaded to many virus forums and the kind folks there will help you out.

3. Download, Install and Run Malwarebytes
Malwarebytes Anti-Malware is an excellent program for finding and removing those really pesky programs that can keep your virus software from doing its job.

  • Download and install Malwarebytes (There is a fee version)
  • Update (just like with your virus program, an out of date malware database is of no use).
  • Run the program
  • Follow the prompts
  • When the scan is finished (This could take several minutes).
  • Click ?See Results?. You should see a list of possible malware. Check these to see that Malwarebytes hasn?t misidentified a useful program for a bad one
  • Click ?Remove Selected?

4. Launch and update your Virus Program.
Always update the program before scanning.? New viruses appear all of the time.? A scan with an outdated virus database is not a complete scan.

  • Scan your computer.?
  • Reboot the computer
  • Scan the computer again

5. Download, Install, and Run a Spyware Program

Again, there are several good spyware programs out there.? Many are free.? My personal favorite at the time of this writing is Spybot.?
One note about Spybot, it also comes with an optional add-on called TeaTimer that helps to protect you from future infections.? As good a program as this is, I find that it can often take up valuable resources while running in the background.? When I install Spybot, I decline the additional install of TeaTimer.
Follow the same procedures as above when running your spyware scan. Reboot, go into Safe mode, update, scan, reboot, and scan again.

6.?Download, Install, and Run an Adware Program
Adware is not as common a problem as it used to be.? I think many advertisers have found that this tends to alienate potential customers; therefore they don?t do it as often anymore.? Also, most Adware is caught by a good spyware program such as Spybot.? When I used to run a lot of adware scan, I would use Adaware.? But I haven?t had to use it on years.

7. System Restore
I know people who swear by this method but I am always a little leery of losing data, it?s never happened but that?s my thoughts, so I use this as a last resort.?
Windows contains a System Restore feature that allows you to rollback you computer to the status it was in before any tragic problems.? To find System Restore

  • Click the Start button
  • Click ?All Programs?
  • Click on the ?Accessories? folder
  • Click on the ?System Tools? folder
  • Click ?System Restore?

System Restore will ask you if you want to restore your computer to an earlier date.? Follow the instructions. Microsoft has made this extremely simple.? Pick a date that you know is prior to your infection and allow Windows to restore your computer to that date.? The computer will restart during this process.?

8. Really tough stuff
The procedures above tend to get things back in order most of the time.? However,? if you are still having troubles there is a possibility that you have a rootkit, a program which reloads itself every time you boot, and if your virus and spyware scans cannot seem to find and root out (pun intended) the small file that allows this to happen, then you will continue to experience virus symptoms. The solution to this is often more drastic in nature and more risky and I will not go into it here. This is when you get out of the ?layman? category and start becoming an expert. If you are looking for more information on these issues, begin by looking for the following softwares. A word of caution, though, before using them read everything you can about them. Read forums and articles from other people who have used them and proceed carefully.?

Two Palestinians killed in Israel-Gaza violence surge

GAZA: An Israeli air strike killed two Palestinians in the Gaza Strip on Tuesday near the border with Israel and the enclave's ruling Hamas movement fired rockets across the frontier for the first time in more than a year.

Palestinian medical officials said the two men were civilians. The Israeli military said aircraft "targeted a terrorist squad identified handling an explosive device" in the central Gaza Strip near Israel's border fence.

The Islamist Hamas group said its Izz el-Deen al-Qassam brigades fired four rockets into Israel in response. No casualties were reported.

Although other militant groups have launched rockets across the border in previous surges of violence, Hamas had held its fire under unofficial truces with Israel.

On Monday, Israeli air strikes killed four Palestinian militants in the enclave, attacks which Fawzi Barhoum, a Hamas spokesman, said moved the group's military wing "to take a firm stance" and launch rockets.

Hours before the aircraft went into action on Monday, militants who crossed into Israel from Egypt's Sinai desert fired on Israelis building a barrier on that frontier, killing one worker, before soldiers shot dead two of the attackers.

The Sinai attack, launched soon after the Muslim Brotherhood declared victory in Egypt's presidential election, raised Israeli concerns about lawlessness in the area since the fall of president Hosni Mubarak last year.

Why Microsoft has to build its own tablet

Microsoft has to do it. That's what I keep telling myself. In order to convert people from the cloud-powered "ecosystems" of Apple or Google, in order to carry Windows desktop users into the post-desktop age, Microsoft needed to introduce a product proudly emblazoned with the Windows logo, something over which it has complete design control: The perfect Windows 8 tablet.

At first look (and feel), the Microsoft Surface tablet is a solid product aimed?in the right direction. Sure, it's a tablet that, from five feet away, looks like all the rest. But it's got a premium magnesium alloy build, a screen that reduces glare and improves interaction by reducing the layers between the LCD itself and the viewer's eye, and clever features like a flush built-in kickstand.

Microsoft even one-upped Apple in a way?by adding a multi-touch keyboard to its own magnetically connected case, the Touch Cover.?Microsoft's tablet?pitch has long been, "Yes, you can touch it but sometimes you need a mouse, keyboard or stylus." At least in the keyboard category, Microsoft is now?approaching the challenge with innovative thinking and a touch of class.

But for all the innovation and design smarts of the new tablet, there's no guarantee it will be a hit.

There are still a lot of unanswered questions:

  • What are the exact?processor and display specifications, not to mention the price, of the ARM tablet?
  • What kind of battery life will it deliver?
  • Will it run on a 3G/4G cellular network?
  • What kind of compatibility will there be with Xbox and PC games?
  • And, most importantly, how will the salesperson at Best Buy answer the question: "Why should I buy this, instead of the Asus, or the Samsung, or the Dell?"

There are few hardware companies that aren't in an utter state of flux (reflux?) over the changing landscape. While Apple designs and markets?both the hardware and the software itself, other companies share the work.?Microsoft would release an?operating system and hardware makers would build products that used it. Google has had reasonable success?using the same approach with Android. But?the shared approach won't last much longer.

That's because there's now?this thing called the "ecosystem." Giants like Apple, Google and Microsoft are locking down customers by providing streaming music, video rental,?cloud storage, email, calendars and all kinds of other services that are exclusive to their products. The more the?ecosystem grows, the more control the giant needs over all of the products. ?

Microsoft tried a middle approach with Windows Phone. It let hardware partners build the devices, but it applied strict specifications. The result was not a success. In fact, it led to confusion: Some phones had beautiful (but small) OLED screens; others had huge (but ugly) LCDs.?One phone streamed music and video to your home theater; another had a kickstand for watching movies on airplanes.

And when Nokia came along with its Windows Phone?flagship, there was no clarifying moment, like when Verizon introduced the Motorola Droid and all of a sudden, Android made sense.?No, the Nokia Windows Phone was just another phone ? one that didn't run Android or iOS.

But even if?the whole hardware-software partnership thing no longer works, how sure can we be that?Microsoft is?prepared to pull an Apple, and go it alone?

After all,?Microsoft's hardware history is hardly a trail of gold. The company has built mice since time immemorial, but they're usually not as lauded as Logitech's. The Xbox is a hot selling console, and rightfully so, but any gamer over the age of 10 can recall the "Red Ring of Death" era, where the percentage of dud systems was remarkably high. Maybe the least besmirched of all Microsoft hardware was the Zune HD, a very nice product that sold ... not very nicely.

And Microsoft has backed out of a few hardware enterprises that turned out to be bad for business: There were the wireless routers, there was Web TV, there was the Kin.

I am not trying to pour salt on wounds that have only recently healed. I want to emphasize a key question: Why did Microsoft embark on this new hardware venture when it has so often failed in the past?

Because it has to.

Unfortunately, once you've swallowed that, there's another?draught you have to gulp down: Windows 8.

I don't share in the ire that some of my colleagues have against Windows 8. However easy or difficult it is to use, its overarching problem is that it is not "Windows" as we've known it through the years.

Windows 7 is Windows. Windows 8 has the same name, but a whole new set of programs, behaviors and metaphors. Calling it, I don't know, "Tiles," instead of "Windows" might have helped, but it would have been an admission of the challenge ahead. Rather than being an upgrade of the tried and true OS, it's a brand new finger-friendly tablet OS, one that will have to get in line for paying customers?behind iOS and Android.

Still, the analysts say that within a few years Microsoft will gain serious headway in the global smartphone market, and that much of this success is due to the fact that it will sell Windows 8 with the same briskness that it has of late sold Windows 7. If you buy the PC or the tablet, you'll learn to love the interface, and you'll buy the phone ? or vice versa.

So how will Microsoft get people to buy the new Windows 8?PCs and tablets? By waiting for Dell or Samsung or Asus or HP to get out there and shove them in people's faces? No.?

It will do it?by handling the whole operation internally, and controlling the message. Microsoft must somehow?reassure?current Windows users (over 1 billion worldwide) that Windows?8 is not a leap of faith but a step forward. And the company must reach out to new audiences with the promise of an ecosystem that's?potentially more compelling than Apple's or Google's, one that ties together home theater, laptops, phones and tablets, and offers a ton of cloud services and apps.?

Microsoft will put its hardware-partner?relationships at risk, but it can afford to, and really it must, if it intends to reaffirm its position of leadership in the tech world.

Karl Gude: Five Reasons Why Twitter Is Amazing In Large Lecture Classes

The guest professor in my large class of 200 journalism students at Michigan State University was just hitting his stride when suddenly every single student plunged to the ground as though looking for a dropped pencil. Stunned, the speaker continued his talk. About 15 minutes later the students leapt to their feet and applauded furiously! Shattered, he began to realize that something he was saying, some word, was igniting this explosive response from the students.

At the time I was on a flight to Denver and the speaker was doing me a favor taking over my class. As the plane was about to land I got a text message from him (yes, I had my phone illegally turned on) that simply said, "You're dead!"

He was right to blame me ... and Twitter.

Now, I wouldn't advise doing this to just any old professor. This was a good friend of mine and I knew he would appreciate a Gude joke. The day before class I had tweeted my students a couple of times encouraging them to commit these outrageous acts whenever my friend spoke a certain word.

Now why would I do such a mean thing? Usually, when a class has a substitute teacher, students just ditch it. Or if they do come to class, they ignore the speaker and spend their time on Facebook. But this little joke caused them to not only attend class (who wouldn't want to miss the fun?) but also to listen intently to every single word the speaker said. Mission accomplished: class was packed and they did well on the quiz I gave later on the material.

More and more students have Twitter accounts now. Four years ago very few of them did and I would have to force them kicking and screaming to sign up (something they were always grateful for by semester's end).

Five (other) ways I use Twitter in my lecture classes:

1) It gives voice to the shy: Normally when I ask a question in a large class the same eager six students (sitting up front) answer them all semester. But, if you tell your class to tweet their answer to the class hashtag and then project the twitter feed onto the big screen, it's overwhelming the number of students who answer the question. I will then pick through them and students can view the ones I don't discuss later. They keep their answers pretty serious, too, since their answer is tied to their identity, but there is always the occasional joke, which we all chuckle over. Obviously, any student who wants to answer in person is happily called on!

2) Students can get answers from their classmates, not just me: Normally, if a student wants to clarify when an assignment is due, they would email me and I would respond. But with Twitter they tweet the question to the class hashtag and any student can answer (often before I do) and this benefits other students who might be wondering the same thing. All boats rise with the tide.

3) Students can only ask a short question and I can only give a short answer: In an email, students will ramble on and on explaining something like why they can't make it to class ("My brother's-friends-uncle's golden retriever that's been in the family for 12 years needs to be rushed to the vet for emergency bypass surgery and I'm the only one who has a car..."), but with Twitter they can only write 140 characters and I can only respond with 140 characters. Nice. Fast. Easy. Of course, if there is something too personal for Twitter, they email me. (No direct messages (DMs) from them since I don't follow them back. Too many!)

4) It's terrific for organizing group projects: Students can use the class hashtag to assemble a group to work on a class project. Then, they can create their own group hashtag for communicating with each other. I then ask them to post their hashtags on the class wiki which allows me to monitor their progress.

5) Students can share relevant links: Twitter is great for sharing links, and students love to share cool discoveries with each other. These links can be collected on a class wiki as a resource.

City?s graduation rate flat for Class of 2011; Just 21% leave high school on time and prepared for college

For the first time in more than five years, the city?s graduation rate remained flat, with 61% of students graduating from high school on time.

At the same time, just over 1 in 5 of the city?s Class of 2011 were considered prepared for college by last June, state figures released Monday show.

State officials trumpeted the increase in graduation rates statewide, but warned that further improvements are needed.

?Too many of those students who do graduate aren't ready for college and careers,? said state Regents Chancellor Merryl Tisch.

"These numbers make clear that we need to continue to pursue aggressive reforms in our schools including a new, richer curriculum and implementation of the new teacher evaluation law in districts across the state."

City officials ignored the bad news, trumpeting the increase in the graduation rate for students who completed high school by August of last year.

?The Class of 2011 can smile again today as we recognize their accomplishments and wish them luck in college and careers,? said Schools Chancellor Dennis Walcott.

?We must do everything we can keep our progress going, and that means continuing to create new schools and improve the quality of teaching in our classrooms.?

(AP) ? The federal and state probes into former Red Sox pitcher Curt Schilling's video gaming company, which filed for bankruptcy protection this week, extend to all the firm's financial transactions, the head of the Rhode Island state police said Friday.

Col. Steven O'Donnell said state police, the state attorney general, the U.S. attorney in Rhode Island and the FBI are conducting the investigation into 38 Studios. He didn't elaborate.

The U.S. attorney's office confirmed the investigation but would not provide details.

The Providence-based company, along with a sister operation in Baltimore, filed for bankruptcy Thursday in Delaware.

The parent firm owes $150.7 million and listed its assets at $21.7 million, according to court filings. 38 Studios Baltimore owes more than $121.4 million and has assets of more than $335,000, the filings show.

In both filings, the company lists its biggest liability as $115.9 million in debt from bonds backed by the state, interest on the bonds and fees to Rhode Island.

Schilling, who also pitched for Baltimore, Houston, Philadelphia and Arizona and won the World Series three times, owns 83 percent of the company, according to the filings.

38 Studios moved to Providence from Massachusetts in 2010 after the state's economic development agency approved a $75 million loan guarantee that was supposed to lead to hundreds of jobs and millions of dollars in tax revenue.

Now, with the firm's bankruptcy filing, state officials are trying to determine how much taxpayers might be on the hook for.

Gov. Lincoln Chafee, who vigorously opposed the loan guarantee on the campaign trail, said Thursday he would do everything possible to salvage what he could of the state's investment.

"If there's a penny that we can get, or a nickel or a dime, we'll get it," he said.

In a statement, 38 Studios said it tried for weeks to find a way to stay afloat but concluded bankruptcy was the only option.

"After ongoing negotiations with the state of Rhode Island and potential investors and other interested parties, the company has been unable to find a solution to the current stalemate," the statement said.

A message seeking comment was left for Schilling on Friday.

Last month, the company laid off nearly 300 employees in Providence and more in Maryland. The move came after it was more than two weeks late on a $1.1 million payment to the state.

38 Studios has been seeking millions of dollars in tax credits from Rhode Island as it struggled to stay solvent, a request that led to accusations the company was looking for a bailout.

As a condition of the loan guarantee, 38 Studios granted Rhode Island a security interest in all its assets.

Chafee said Thursday that the filing wasn't a surprise. The state recently selected an outside auditor to review the finances of Schilling's company. Chafee had indicated he didn't want to give the company any more public support at the risk of throwing "good money after bad." Instead, the state tried to help find private investment.

"We helped as much as we could," he said.

38 Studios owes at least $2.5 million to employees in Rhode Island and Maryland and $322,000 in federal and state taxes, including $39,000 to Rhode Island, according to the bankruptcy filings.

It owes fantasy writer R.A. Salvatore, who helped develop Schilling's first game, "Kingdoms of Amalur: Reckoning," more than $1.7 million. The game came out to strong reviews in February; Schilling said it beat sales expectations, but Chafee has called it a "flop."

The parent company listed no income from its business operations from Jan. 1, 2010, through May 31 of this year. The Baltimore company listed $22 million in income under a "work for hire agreement" with Electronic Arts Publishing, which published "Kingdoms of Amalur," and $635,000 in royalties during the same period.

Schilling wasn't drawing a salary, the filings show. The bankruptcy filing names Schilling as a co-debtor and lists two personal guarantees he made on credit lines totaling more than $3.5 million.

Chief Executive Officer Jen MacLean was paid $253,000 a year, while President and Chief Operating Officer William Thomas made about $200,000.

DETROIT - A Wayne State University researcher believes a protein that fails to reach the nucleus of retinal cells may play a role in causing eye disease in people with diabetes.

Renu A. Kowluru, Ph.D., professor of ophthalmology, anatomy/cell biology and endocrinology at Wayne State University and the Kresge Eye Institute, recently received a one-year, $110,000 Innovative Grant from the Juvenile Diabetes Research Foundation to help gain new insight into the development of diabetic retinopathy and identify targets for future therapeutic interventions.

Her hypothesis is that NF-E2-related factor 2 (Nrf2), a protein that regulates antioxidant response, fails to reach to the nucleus of cells in the retina to neutralize free radicals, which are volatile. As a result, Kowluru said, KEAP1 - the protein that serves as an anchor for Nrf2 and represses its activation - increases, damaging the mitochondria, the cells' power source. That damage, she believes, in turn accelerates the death of cells in tiny blood vessels (capillaries), ultimately resulting in the development of retinopathy.

Diabetes is a leading cause of acquired blindness in young adults. Almost 50 percent of diabetes patients have some form of retinopathy - not necessarily blindness - after nine years, and that figure increases to 95 percent after 20 years of the disease.

"Basically it doesn't leave any diabetic patient," Kowluru said.

Titled "Role of Nrf2-KEAP1 in Diabetic Retinopathy," her project will look at the oxidative stress, an imbalance between the production and utilization of free radicals, in an effort to determine how a disease state - diabetes in this case - causes that imbalance.

"We have shown that in diabetes, mitochondria are damaged, and when they are damaged they initiate cell death," Kowluru said. "We want to see what the role of Nrf2 is in initiating this signal."

Beginning in October, her team will genetically regulate Nrf2 to examine the temporal relationship between the failure of the protein to reach cells' nuclei and the signal sent by damaged mitochondria that triggers cell death. She hopes to determine if the former event precedes the latter, or if they occur at the same time.

Additionally, Kowluru will examine the point on the gene where Nrf2 and KEAP1 interact to determine whether diabetes induces epigenetic changes in Nrf2, impairing its binding with KEAP1.

Because epigenetic changes, which are functionally relevant modifications to the genome that do not involve a change in the DNA sequence, can be passed along to future generations, prevention becomes an even more important goal, she said. Drugs now in the clinical trial stage for other chronic diseases (e.g., cancer) may be able to prevent epigenetic changes in the retina.

"Once we understand the problem, we can administer supplements and prevent the downstream pathway for retinopathy," Kowluru said.
